In vitro Callus Formation and Shooting Responses of Jujube as Affected by Growth Regulators and Levels of Inorganic Nutrients





Abstract
Node-bud segments of jujube (Zizyphus jujuba Mill.), prepared from subcultured explants, were cultured in vitro on basal MS media supplemented with various concentrations of growth regulators and inorganic nutrients.
The rate of callus formation was high on the media supplemented with either 0.5§·/§¤ 2, 4-D or 0.5§·/§¤ picloram, while shoot development was promoted by 0.5§·/§¤ 2, 4-D.
Callus growth, fresh weight, and the survival rate decreased as the levels of casein hydrolysate increased.
1.0§·/§¤ kinetin was effective in the shoot development, and the callus formation was high on the media supplemented with either 1.0§·/§¤ BA or 0.5¡­1.0§·/§¤ 2iP.
Higher rate of callus formation was obtained with half strength MS medium supplemented with 0.5§·/§¤ 2, 4-D, while half and full strength MS media supplemented with 1.0§·/§¤ IBA+1.0§·/§¤ BA were more favorable for shoot growth.
Organogenesis was inhibited and delayed .as the levels of NH©þNO3, KNO3, and KH©üPO©þ were lower than the recommended rate of MS medium.
